摘要
[目的]探讨野外试验的影响因素。[方法]在塔里木河下游典型断面阿拉干选择胡杨树,分3层采集胡杨叶片,测量其相对含水量( FMC)、等效水厚度( EWT)和叶绿素含量,探讨胡杨不同层次叶片含水量的差异以及代表胡杨叶片平均含水量和叶绿素含量状况的适宜采样量,并分析胡杨叶片一天中含水量和叶绿素含量的变化状况。[结果]千分之一精度天平即能满足测量一片叶片重量需求,冰袋保鲜的时长能够达到野外试验的要求;胡杨上、中、下不同层次的叶片含水量之间有显著差异( P<0.01),采集15片叶片与20片叶片时FMC、EWT变化率较小,所以采样时需分3层采样,每层采集15片叶片,叶片尺寸要求分布均匀;使用SPAD-502测量胡杨叶片叶绿素含量,每棵树采集5、10、15片时,没有显著差异(P>0.05),即测量5片即可;一天中叶片FMC、EWT变化较大,但叶绿素含量基本不变化(1.5SPAD单位内)。[结论]该研究结果可为胡杨的野外试验研究提供参考,同时也为大范围反演胡杨生理状况研究提供试验研究。
Abstract
Objective] The aim was to discuss influencing factors of field experiment.[Method] Selecting Populus euphratica from typical section Arakan in the lower reaches of the Tarim River, collecting leaves from 3layer, the FMC, EWT and chlorophyll content were meas-ured, differences of leaf moisture content at various levels were discussed , as well as the optimal sampling quantity which can represent aver-age leaf moisture content and chlorophyll content, the change of leaf moisture content and chlorophyll content within a day was analyzed.[Re-sult] Libra over one thousand scale precision could satisfy the demand of measuring a piece of leaf weight , ice bag preservation duration could meet the demands of field experiments; there was significant difference in leaf moisture content ( P<0.01 ) .FMC and EWT had low rate of changes between 15 and 20 leave samples: we should sample leaves from 3 leaf layers and 15 leaves per layer.There were insignificant( P>0.05) correlation between 5, 10 and 15 leaves, we just need to measure 5 leaves as chlorophyll content.FMC and EWT fluctuated within a day, while the chlorophyll content remained stable(within 1.5 SPAD).[Conclusion] The study can provide reference for field experiment of Populus euphraitca, also provide research on large-scale inversion of physiological state of Populus euphratica.
