摘要

采用离心后不同的低温保存方式和添加新鲜培养基的方法,对角毛藻离心收集后储存的最长时间进行了研究。结果表明,添加新鲜培养基的储存方法优于直接添加藻液的方法,并且存储时间长;角毛藻在3000 r/min或较低转数离心后,藻泥可以添加新鲜培养基储存于-2℃冰箱,至少储存35 d,再培养不影响生长。角毛藻储存温度越低,储存时间就越长,但要在角毛藻不结冰的情况下。此保藏方法可以应用于角毛藻销售中转储存。

Abstract

After centrifugation, different low-temperature storage method and the method of adding fresh culture medium were applied to re-search the longest storage time of Chaetoceros muelleri.Results showed that the storage method of adding fresh culture medium was superior to directly adding algae liquid.After algal biomass was centrifuged at speed of 3 000 r/min or lower speed, fresh medium was added into the sludge and stored at -2 ℃ in refrigerator for at least 35 d.Subculture did not affect the growth.The lower storage temperature of C.muelleri made longer storage time under the condition of non-freezing.This storage method could be used for transitional storage in the sale of C.muel-leri.
