摘要
[目的]研究山葡萄嫩茎段愈伤组织离体培养最佳配方。[方法]以山葡萄优良母株新生枝条嫩梢为外植体,观察山葡萄在IAA和NAA不同浓度下芽的分化、继代与生根情况。[结果]在相同条件下,不同浓度IAA和NAA对山葡萄的芽分化、继代与生根影响显著。芽分化NAA浓度为0.05 mg/L的时候效果最好,愈伤组织中等,分化出的芽较多。继代IAA浓度为0.10 mg/L的时候效果最好,生长旺盛,繁殖倍数较高。生根NAA浓度为0.10 mg/L的时候效果最好,根粗壮,生根率高。[结论]该研究结果可为山葡萄离体培养奠定基础。
Abstract
Objective] To discuss the optimum formula for the in vitro culture of tender stem callus of Vitis amurensis Rupr.[ Method] New-born branch tops of good maternal plants of V.amurensis were used as explants, and the bud differentiation, subculture and root growth of V. amurensis were analyzed under different concentrations of IAA and NAA.[Result] Under the same conditions, different concentrations of IAA and NAA had obvious effects on the bud differentiation, subculture and root growth of V.amurensis.When NAA concentration was 0.05 mg/L, callus was moderate, and there were more buds.As IAA concentration was 0.10 mg/L, it grew well, and the breeding rate was high. When NAA concentration was 0.10 mg/L, its roots were thick, and the rooting rate was high.[Conclusion] The study can lay the foundation for the in vitro culture of V.amurensis.
