摘要

[目的]研究发酵土鳖虫含水醇提物中总黄酮含量的测定方法,并对其方法学进行考察。[方法]采用NaNO2-AlCl3-NaOH比色法在510 nm处测定黄酮类化合物与显色剂所形成的络合物吸光度。[结果]所测定标准曲线线性相关性良好,r=0.9990,线性范围在0.02~0.10 mg/mL,试验方法的精密度RSD=1.92%,重现性RSD=1.23%,样品回收率95.00%±10.00%,RSD=2.09%。样品重复测定总黄酮含量平均值为4.6308 mg/g。[结论]方法学考察结果表明,用紫外分光光度法测定发酵土鳖虫总黄酮类化合物含量的标准曲线和线性范围、精密度、准确度和重复性等项内容都符合相关规定,该方法准确可靠,可用于含量测定。

Abstract

Objective] To detect the total flavonoids content in ethanol extracts from fermented Eupolyphaga sinesis Walker, and to investigate the methodology.[ Method] NaNO2-AlCl3-NaOH colorimetry was used to detect the complex absorbance formed by flavonoids compounds and color developing agent.[Result] The detected standard curve had good linear correlation, r =0.999 0.The linear range was between 0.02 and 0.10 mg/mL.The precision of the test was RSD =1.92%; repeatability was RSD =1.23%; sample recovery rate was 95.00% ± 10.00%, RSD=2.09%.The average content of total flavonoids was 4.630 8 mg/g.[ Conclusion] Results of methodological investigation showed that the standard curve, linear range, precision, accuracy and repeatability of total flavonoids content in fermented E.sinesis all meet the requirements of relevant provisions, which are detected by ultraviolet spectrophotometry.This method is accurate, reliable and can be used for content detection of total flavonoids.
