摘要

[目的]利用离子交换层析方法分离与纯化口蹄疫病毒抗原。[方法]利用阴离子交换树脂制备高纯度的FMDV抗原,通过对样品及缓冲液pH、流速、缓冲液的盐浓度、样品上样体积等条件的摸索,确定最佳的FMDV抗原洗脱条件。[结果]当缓冲液pH为8.0、流速1.2 mL/min、盐浓度450 mmol/L、上样体积1.5 mL时,FMDV抗原的纯化效果最佳。[结论]该研究结果可为离子交换层析在FMDV抗原的分离与纯化提供依据。

Abstract

Objective] The aim was to separate and purify FMDV antigen by using ion exchange chromatography.[Method] High purity FM-DV antigen was prepared by anion exchange resin, through exploration on sample, buffer solution pH, current speed, salt concentration, load-ing volume of sample, the optimal elution condition of FMDV antigen was determined.[ Result] The best condition for purification of FMDV antigen is when buffer pH reached 8.0, flowed at a speed of about 1.2 mL/min, salt concentration was 450 mmol/L and the loading volume of sample was 1.5 mL.[Conclusion] The results can provide basis for separation and purification of FMDV antigen by ion exchange chroma-tography.
