摘要
[目的]提高克隆猪的大批量生产效率,优化体外与体内培养体系。[方法]将所获得的猪体细胞克隆胚胎用含不同Vc浓度的培养液培养,比较体外囊胚发育力和囊胚细胞数,筛选出合适的培养浓度,然后用此浓度培养后的胚胎进行体内手术移植,统计克隆猪的分娩率和产仔情况。[结果]克隆胚胎在激活后用40μg/mL Vc处理22 h,体外培养未显著增加囊胚率,但显著提高了囊胚细胞数。体内移植结果显示,添加Vc未提高妊娠率,但增加了窝均总仔数,克隆效率差异显著。[结论] Vc处理有利于增强体细胞克隆胚的体内和体外发育能力。
Abstract
Objective] To improve the ability to generate cloned pigs and to optimize the culture system in vitro and in vivo.[Method] The por-cine SCNT embryos were cultured in the medium with different vitamin C concentrations .Then the blastocyst formation rate and cell number per blastocyst were compared.Proper concentration was screened,which was used to treat cloned embryos.After surgery transfer,farrowing rate and litter size were calculated.[Result] After stimulation,cloned embryos were treated by 40μg/mL Vc for 22 h;culture in vitro did not significantly enhance the blastocyst rate,but the blastocyst cell number was significantly increased.Results of in vivo test showed that adding Vc did not signifi-cantly enhance the pregnancy rate,but increased the average litter size.There were significant differences in the cloning efficiency.[Conclusion] Vc treatment is beneficial to the development of porcine SCNT embryos both in vivo and in vitro.
