摘要

[目的]采用苯酚硫酸法和蒽酮硫酸法测定托盘根多糖含量。[方法]用精制托盘根多糖测得对葡萄糖的换算因子,通过线性关系、精密度、重现性、稳定性及加样回收率等指标进行综合评价。[结果]葡萄糖在5~40μg/mL范围内浓度与吸光度呈良好的线性关系,重复性试验相对标准偏差<1.7%,加样回收率≥97%,苯酚硫酸法和蒽酮硫酸法测定的多糖含量分别为3.78%和4.02%。[结论]苯酚硫酸法和蒽酮硫酸法均准确可靠,适合于托盘根多糖的测定。

Abstract

Objective] The phenol-sulfuric acid method and anthrone-sulfuric acid method were compared on determination of the polysac-charide content in Rubus crataegiflolius Bunge..[Method] The conversion factor was measured with refined polysaccharide for glucose, and comprehensive evaluation was carried out by linear relationship, precision, reproducibility, stability and recovery rate.[ Result] The results showed that the linear relationship between glucose concentration and absorbance was good in the range of 5-40 μg/mL, and the average recov-ery rate was equal to or higher than 97% with good reproducibility (RSD=1.7%), the content of polysaccharide determined by phenol-sulfu-ric acid method and anthrone-sulfuric acid method were 3.78% and 4.02% .[ Conclusion] Phenol-sulfuric acid and anthrone-sulfuric acid methods are accurate and reliable, and suitable for determination of polysaccharide in R.crataegiflolius.
