摘要

[目的]以京尼平苷为原料,在自然pH下,通过产β-葡萄糖苷酶的邬衣黑曲霉发酵将京尼平苷转化为京尼平。[方法]通过单因素和正交试验考察了京尼平苷液浓度、发酵温度、发酵时间、邬衣黑曲霉孢子的接入量、摇瓶装量对京尼平生成量的影响。[结果]最终确定最佳工艺为:在自然pH条件下,以0.20%京尼平苷液为发酵培养基,按菌株孢子与发酵培养基3.0∶10.0(V/V)的比例接入邬衣黑曲霉孢子,摇瓶装量为100 mL/250 mL,28℃,200 r/min发酵66 h。发酵结束后高效液相色谱检测京尼平,结果显示京尼平苷的转化率可达99%以上,转化产物京尼平纯度可达98%以上。[结论]该研究所用方法安全、高效,是生产京尼平的可行方法。

Abstract

Objective] To transform geniposide to genipin by fermentation of Aspergillus usamii that can produce β-glucosidase under the con-dition of natural pH.[Method] Through a single factor experiment and orthogonal design experiment, the effects of geniposide concentration, fermentation temperature, fermentation time, inoculation quantity of A.usamii spores, and bottled liquid volume on the production of genipin were analyzed.[Result] The optimum process is shown as follows:under the condition of natural pH, 0.20%geniposide was as the fermenta-tion medium, and the proportion of the spores to the fermentation medium was 3.0∶10.0 (V/V);bottled liquid volume was 100 mL/250 mL;the fermentation was conducted for 66 h at 28 ℃ at the rotating speed of 200 r/min.Under the optimum conditions, the purity of genipin was more than 98% ,and the conversion rate of geniposide reached more than 99% through HPLC testing.[ Conclusion] The method is safe, effi-cient and feasible to produce genipin.
