摘要

[目的]探究产淀粉酶菌种的分离纯化及培养条件。[方法]从土壤中分离到1株产淀粉酶能力较强的细菌A并分离纯化培养。[结果]细菌A在以淀粉为碳源的培养基上产淀粉酶能力最强,水解圈平均直径/菌种平均直径(R2/R1)为2.69;在以硝酸钠为氮源的培养基上产淀粉酶能力最强,R2/R1为2.69;pH在7.5时产淀粉酶能力最强,R2/R1为2.95。[结论]以淀粉为碳源、以硝酸钠为氮源、 pH在7.5的培养基有利于细菌A产淀粉酶能力的提高。

Abstract

Objective] To explore the separation, purification and culture conditions of an amylase producing strain.[Method] Bacterium A, which had a strong ability to produce amylase, was isolated from the soil and then purified and cultured.[ Result] Bacterium A had the stron-gest ability to produce amylase in the medium using starch as the carbon source, and the ratio of the average diameter of hydrolyzation circles R2 to the average diameter of the strain R1(called R2/R1 for short) was 2.69;when being cultured in the medium using sodium nitrate as the nitrogen source, bacterium A had the strongest ability to produce amylase, and R2/R1 was 2.69;as the pH of the medium was 7.5, the ability to produce amylase was the strongest, and R2/R1 was 2.95.[Conclusion] Bacterium A should be cultured in the medium where starch and so-dium nitrate are as the carbon source and the nitrogen source and pH is 7.5 to improve the ability to produce amylase.
