摘要
[目的]研究培养基中葡萄糖、蛋白胨、丙氨酸对黑木耳锁状联合菌丝比率的影响.[方法]采用平板插片培养方法,以黑木耳H29为试验材料,通过单因素试验确定葡萄糖、蛋白胨、丙氨酸对锁状联合菌丝比率促进的最佳浓度.再通过正交试验确定其最佳组合.[结果]适当浓度的葡萄糖、蛋白胨、丙氨酸均能提高H29锁状联合菌丝比率,其最佳组合为20 mg/mL葡萄糖、6mg/mL蛋白胨、3.5 mg/mL丙氨酸.[结论]该研究为黑木耳的育种和实际生产提供参考.
Abstract
[Objective] To study the effects of glucose,peptone and alanine in culture medium on clamp-connection hyphae ratio of Aurieuzaria auricular.[Method] With A.auricular H29 as the test materials,plate culture method was adopted.The optimal concentrations of glucose,peptone and alanine that promoting the clamp-connection hyphae ratio were determined by single factor test.The optimal constitution was determined by orthogonal test.[Result] The appropriate concentrations of glucose,peptone and alanine could improve the clamp-connection hyphae ratio of A.auricular H29.The optimal concentration combination was 20 mg/mL glucose,6 mg/mL peptone and 3.5 mg/mL alanine.[Conclusion] This research provides references for the breeding and practical production of A.auricular.
