摘要
[目的]筛选对镉有高耐性的菌株,为探究重金属污染微生物修复技术提供理论依据.[方法]以DN-1和DN-2为供试菌株,在不同培养时间、培养温度、pH、镉离子初始浓度的条件下,研究不同菌株对镉离子的吸附效果.[结果]培养3d时菌株DN-1和DN-2对镉离子的吸附率最大,分别为61.92%和89.57%;2种菌株对镉离子的最佳吸附温度为30℃.菌株DN-1和DN-2分别在pH为8和6时吸附率最高;镉离子初始浓度为80~200 mg/L时,DN-2吸附率明显高于DN-1.透射电镜观察发现,与未经处理的菌株相比,经镉离子处理后的菌株细胞表面及胞内有沉淀物聚集,细胞内部结构发生明显变化.[结论]菌株DN-1和DN-2对镉离子具有一定耐受性,吸附效果明显.胞外吸附和胞内沉淀可能是菌株吸附重金属的重要途径.
Abstract
[Objective] The aim was to screen out strain with high tolerance to cadium,to provide theoretical basis for remediation technology of heavy metal contaminated microorganism.[Method] With DN-1 and DN-2 as tested strains,under different conditions of culture time,temperature,pH,initial concentration ofCd2+ solution,the effect of different strains on the adsorption ofCd2+ was studied.[Result] The results showed that under certain experimental conditions,when cultured for three days the adsorption rate of DN-1 and DN-2 to Cd2+ was the highest,which were 61.92% and 89.57%,respectively;The optimum adsorption temperature was 30 ℃.The optimum pH of DN-1 and DN-2 was 8 and 6;When the initial concentration of Cd2 + was 80-200 mg/L range,DN-2 adsorption rate was significantly higher than that of DN-1.Through the transmission electron microscope observation can found that after Cd2+ treatment,the cell surface and intracellular accumulation of sediment compared with untreated strains,the internal structure of cells changed obviously.[Conclusion] DN-1 and DN-2 strains have a certain tolerance to cadmium ions,and the adsorption effect is obvious.Extracellular adsorption and intracellular precipitation may be an important way for the strain to adsorb heavy metals.
