摘要

[目的]建立熟地黄中多糖含量的测定方法.[方法]利用苯酚硫酸法、蒽酮硫酸法、3,5-二硝基水杨酸法(DNS法)分别测定熟地黄中的多糖含量,对检测结果进行比较.[结果]苯酚硫酸法、蒽酮硫酸法、DNS法的平均回收率分别为100.5%、99.8%、100.6%,RSD分别为1.6%、1.6%、1.5%,测得的多糖质量分数分别为14.80、14.66、17.46 mg/g.[结论]苯酚硫酸法的结果更可靠,且操作相对简单易行,可考虑作为熟地黄多糖含量测定的首选方法.

Abstract

[Objective] The research aimed to estabhsh the determination method of polysaccharide content of Radix Rehmanniae Preparata..[Method] The polysaccharide contems of Radix Rehmanniae Preparata.were determined by phenol-sulfuric acid method,anthrone-sulfuric acid method and 3,5-dinitrosalicylic acid method (DNS method) respectively,and the test results were compared.[Result] The average recovery rates of phenol-sulfuric acid method,anthrone-sulfuric acid method and DNS method were 100.5%,99.8% and 100.6%,respectively.The RSD were 1.6%,1.6% and 1.5% respectively.The mass fraction of polysaccharides were 14.80,14.66 and 17.46 mg/g,respectively.[Conclusion] The phenol-sulfuric acid method was more reliable,the operation was relatively simple,which can be considered as the preferred method of determination of oolvsaccharide content of Radix Rehmanniae Preparata.
