福建柏叶片全蛋白3种提取方法的比较

摘要
[目的]探索适于福建柏蛋白质组学研究的全蛋白样品制备的最优方案.[方法]以福建柏叶片为材料,利用3种全蛋白提取方法:TCA/丙酮法、酚提法和TCA/酚提结合法,研究不同提取方法对福建柏蛋白双向电泳结果的影响.[结果]TCA/丙酮法制备的蛋白点数较少,为285个,蛋白质在制备过程中流失较多;酚提法得到的蛋白点最多,为1 226个,蛋白点较清晰但pH中端部分有轻微的蛋白堆积;TCA/酚提结合法得到了813个蛋白点,蛋白点较清晰,无明显的蛋白堆积现象.[结论]在福建柏蛋白质组学研究中,酚提法更适于福建柏全蛋白的制备,且酚提法明显优于TCA/酚提结合法.
Abstract
[Objective] To study the most efficient method for sample preparation of Fokienia hodginsii proteomics.[Method] Three different methods including TCA/acetone,phenol extraction method and TCA/phenol extraction method were analyzed by separation effect of 2-DE with Fokienia hodginsii leaves as material.[Result] The results showed that using TCA/acetone method,285 protein spots could be detected on 2-DE gels which had severest loss in proteins.The phenol extraction method was the best with more clear protein spots as many as 1 226 protein spots on 2-DE gels,but using protocol extraction method,there were slightly protein accumulation phenomena at neutral.TCA/phenol extraction method was produced 813 protein spots on 2-DE gels with more clear protein spots and slight protein accumulation phenomena.[Conclusion] The phenol extraction method is efficient and optimal method for preparation of Fokienia hodginsii total protein,meanwhile the phenol extraction method is better than TCA/phenol extraction method.
