摘要
[目的]建立沅江荻芦嫩茎总氨基酸含量测定方法,研究4种获芦的贮存方法对获芦嫩茎总氨基酸含量的影响.[方法]采用茚三酮比色法测定荻芦中总氨基酸的含量,筛选以谷氨酸为标准溶液的最佳工作条件,在401 nm处测定各样品吸光度.[结果]新鲜荻芦中总氨基酸含量占22.0%,各贮存方式下的总氨基酸含量为4.9% ~21.2%.平均回收率为99.3% ~99.6%,RSD为0.06% ~ 0.52%.[结论]通过比对4种贮存方式下的总氨基酸含量,减压贮存是最有利于减少营养成分氨基酸的贮存方式.该方法简便有效,污染少,可行性好.
Abstract
[Objective] To establish a method for determination of total amino acid content in tender stem of Triarrhena lutarioriparia from Yuanjiang,to study the effects of four storage methods on amino acid content.[Method] Using ninhydrin colorimetry method to determinate total amino acid content in T.lutarioriparia,screen the best working conditions by glutamate as the standard solution,and measure absorbance at its maximum absorption wavelength at 401 nm.[Result] The total content of amino acid in fresh T.lutarioriparia accounted for 22.0%,the amino acid content under each storage method was in 4.9%-21.2%,average recovery was in 99.3%-99.6%,RSD was in 0.06%-0.52%.[Conclusion] By comparing the contents of total amino acids in four ways,reduced pressure storage is the most beneficial way to reduce the amino acid.The method is simple and effective,less pollution and good feasibility.
