摘要

[目的]探讨电刺激对铁皮石斛原球茎中石斛碱含量的影响.[方法]采用铁皮石斛原球茎培养基扩增培养铁皮石斛原球茎,并将扩增所得原球茎随机分为试验组和对照组,其中试验组给予不同时间电刺激(25 V),对照组不做处理,正常培养15 d后,以氯仿热回流法提取原球茎中的石斛碱,紫外分光光度法测定石斛碱含量.[结果]与对照组相比,经电刺激(25 V)1 和2 h的试验组铁皮石斛原球茎石斛碱含量显著增加(P<0.05),而经电刺激(25 V)3 h的试验组铁皮石斛原球茎石斛碱含量下降(P<0.05).[结论]25 V电刺激1、2 h能够促进铁皮石斛原球茎中的石斛碱积累,这可能与电刺激能改变原球茎中细胞的渗透性、增强对培养基中各种成分的吸收,导致内源次生代谢物增加有关.

Abstract

[Objective] To investigate the effects of electrostimulation on dendrobine content in protocorm of Dendrobium officinale.[Method] Protocorms were cultured by protocorm culture medium and randomly divided into two groups, the experimental group was treated with electrostimulation(25 V) while the control group did not be disposed.After 15 days incubation, dendrobine was extracted by chloroform and hot-refluxing, and determinated by ultraviolet spectrophotometry.[Result] Compared with the control group, the dendrobine content of experimental group treated with electrostimulation(25 V) for 1 h or 2 h was increased significantly(P<0.05), while the dendrobine content was decreased(P<0.05) in experimental group treated by electrostimulation(25 V) for 3 h.[Conclusion] The electrostimulation(25 V) for 1 h or 2 h can promote the accumulation of dendrobine in protocorm, and the mechanism may be related to the changed permeability of cell, the enhanced absorption of nutrients and the increased endogenous secondary metabolites.
