摘要

[目的]研究党参毛状根的诱导条件.[方法]利用发根农杆菌A4、C58C1和A1476进行毛状根诱导,探讨发根农杆菌种类、外植体类型、菌液浓度、侵染时间和共培养天数等因素对党参毛状根诱导的影响.[结果]3种发根农杆菌中A4诱导率最高;茎段的诱导率最高;发根农杆菌浓度在OD600为0.4时诱导效果较好;较佳侵染时间为5 min;共培养时间为2 d时诱导率较高;头孢噻肟钠浓度为300 mg/L时除菌效果较好.[结论]该试验为大量生产毛状根以及毛状根的后续研究提供试验基础.

Abstract

[Objective] The research aimed to study the induction condition of hairy roots of Codonopsis pilosula.[Method] The hairy roots were induced by Agrobacterium rhizogenes strains of A4, C58C1 and A1476,the effects of Agrobacterium rhizogenes species, explants types, concentration of bacterium,infection time, co-cultivation period on hairy roots inductivity of Codonopsis pilosula were discussed.[Result] Among the three Agrobacterium rhizogenes,the induction rate of A4 was the highest.The induction rate of stem was the highest.The inductivity at OD600 0.4 was better performing.Better infection time was five minutes.The inductivity was higher when the time of co-cultivation was two days.When the concentration of cefotaxime sodium was 300 mg/L, the sterilization effect was better.[Conclusion] This test provides experimental basis for the mass production of hairy roots and follow-up study of the hairy roots.
