摘要

[目的]考察大孔树脂对黔产野生水芹(简称水芹)水溶性总黄酮的纯化条件.[方法]测定 AB-8、DM-130和HP-20 3种大孔树脂对水芹总黄酮的吸附量及解吸率,筛选树脂型号;以总黄酮含量为指标,采用单因素试验优选HP-20树脂纯化工艺.[结果]HP-20型大孔树脂分离纯化水芹总黄酮最佳的工艺条件为:料液比为1∶30(g∶ mL),静置吸附时间为6 h,解吸液乙醇溶液的体积分数为70%,pH为3,解吸温度为40 ℃,经HP-20处理后的水芹总黄酮纯度较高(提高了9.4倍).[结论]HP-20树脂较适于分离纯化黔产野生水芹水溶性总黄酮,为水芹的开发利用提供科学依据.

Abstract

[Objective] The research aimed to study the purification conditions of water-soluble total flavonoids from wild Oenanthe javanica in Guizhou Province.[Method]The adsorption quantity and desorption rate of three kinds of macroporous resins (AB-8、DM-130 and HP-20) were measured for total flavonoids of Oenanthe javanica,resin type was selected.Using the content of total flavonoids as index,the single factor experiment was used to optimize the purification process of HP-20 resin.[Result]The optimum process conditions of separation and purification of total flavonoids from Oenanthe javanica for HP-20 macroporous resin:solid-liquid ratio was 1∶30 (g∶ mL),standing time was 6 h,the volume fraction of the desorption ethanol solution was 70%,pH value of sample was 3,the desorption temperature was 40 ℃.The total flavonoids had high purity after HP-20 macroporous resin (increased 9.4 times).[Conclusion]HP-20 resin is suitable for separation and purification of water-soluble total flavonoids,to provide a scientific basis for the development and utilization of Oenanthe javanica.
