摘要
[目的]对家兔Myh7基因进行生物信息学分析及表达规律研究.[方法]利用RT-PCR技术检测了Myh7在肌肉发育过程中的表达规律,并结合生物信息学方法对家兔Myh7的序列进行深入分析.[结果]Myh7蛋白为不稳定亲水性蛋白,有Myosin_N和MYSc_class_Ⅱ这2个结构区域,其中MYSc class Ⅱ是肌球蛋白重链家族特有结构域.MEGA软件分析家兔Myh7分子进化,发现其与人、猩猩等物种间同源性较高,与其亲缘关系的远近一致.定量分析显示:在整个肌肉发育过程中,Mhy7表达量在腿肌中基本维持在较低水平,在背肌中的表达量相对波动较大,而在11周龄时出现表达量上升的趋势.[结论]该研究为进一步研究Myh7基因在肌肉生长过程中的功能奠定了分子基础,为研究家兔的肌肉发育分子机制提供了一定的参考依据.
Abstract
[Objective] Bioinformatics analysis and expnession characteristics of Myh7 gene in rabbit were studied.[Method] The expression characteristics of Myh7 was detected by RT-PCR technique in the process of muscle development,and we further used bioinformatics methods to analyze rabbit Myh7 sequence.[Result] Myh7 protein was unstable and hydrophilic,with the Myosin_N and MYSc_class_Ⅱ structure domain,MYSc_ class_Ⅱ was the special structure domain of myosin heavy chain.We used MEGA software to analyze Myh7 molecular evolution of rabbit,the result showed that homology was high among rabbit,human and orangutan,which was conforming to the kinship.Quantitative analysis showed that Mhy7 in leg muscle expressed in the low level,and the expression was fluctuant in the back muscle,increased in 11th week old rabbit.[Conclusion] The study provided the molecular basis for exploring the Myh7 gene function in the process of muscle growth,and it could also provide certain reference basis for studying the molecular mechanism of rabbit muscle development.
