摘要
[目的]建立烟台柴胡中柴胡皂苷a、d含量的HPLC测定方法,研究烟台柴胡中柴胡皂苷a、d的含量,为评价烟台柴胡品质提供科学方法.[方法]采用Thermo ODS-2 HYPERIL(250 mm×4.6 mm,5 μm)色谱柱;流动相为乙腈和水,梯度洗脱;流速1 mL/min;柱温25 ℃;检测波长210 nm.[结果]方法学考察表明柴胡皂苷a、d的峰面积积分值和进样量的线性关系良好,精密度试验的RSD分别为0.67%和0.75%;加样回收率分别为99.4%和98.7%,RSD分别为1.53%和1.76%.不同来源的烟台柴胡中柴胡皂苷a、d的含量相当,总量在0.772%~1.079%.[结论]该方法稳定、重现性好、操作简单,可用于烟台柴胡的质量控制.烟台柴胡中柴胡皂苷a、d含量高于药典规定的柴胡标准,可以作为柴胡的药材新资源.
Abstract
[Objective]The research aimed to provide scientific basis for evaluating the quality of Bupleurum chinese through establishing HPLC method to determine and study the content of saikosaponins a and d in it.[Method]Thermo ODS-2 HYPERIL(250 mm ×4.6 mm,5 μm)column was used. Mobile phase was acetonitrile-water with gradient elution at a flow rate of 1.0 mL/min. Column temperature was 25 ℃ and the detection wave length was 210 nm.[Result]Methodology examination indicated that the linear relation between peak area integral value and saikosaponins a and d sample were good. RSD in precision experiment was 0.67% and 0.75%, respectively. Meanwhile, the respective recovery coefficient was 99.4% and 98.7%,RSD was 1.53% and 1.76%,respectively. There was only a little difference in the content of saikosaponins a and d that exist in Bupleurum chinese from different origins. The total content was between 0.772% and 1.079%.[Conclusion]This method was stable, simple and with good reproducibility, which can be used for the quality control of Bupleurum chinese. Saikosaponins a and d content in Bupleurum chinese is found higher than the standard content of which in Bupleurum chinense according to Chinese Pharmacopoeia, so Bupleurum chinese can be used as a new resource of bupleurum.
