摘要
[目的] 研究不同制剂方式对药物在水产动物体内代谢动力学的影响.[方法]用诺氟沙星粉(NP)、诺氟沙星盐酸小檗碱预混剂(NB)、烟酸诺氟沙星可溶性粉(NN)和乳酸诺氟沙星可溶性粉(NL)4种诺氟沙星制剂,分别混饲口灌日本鳗鲡,给药量均按30 mg/kg 的诺氟沙星原粉计算,再利用超高效液相色谱-质谱法测定诺氟沙星在血浆中的含量,并进行药动学分析.[结果] 非房室模型药动学参数表明,NP、NB、NN和NL 4种诺氟沙星制剂单次给药后均在3 h或4 h达峰,达峰浓度(Cmax)相差较大,分别为1.273、1.073、0.616和0.094 mg/L;消除半衰期(T1/2)分别为15.267、24.887、33.041和25.891 h;NP的AUC最大,为22.670 mg/(L·h),NP:NB:NN:NL=100%:99%:63%:13%.[结论] 药物的不同制剂方式对其在水产动物体内的吸收程度和生物利用度有较大的影响.
Abstract
[Objective] To explore the influences of formulation ways on pharmacokinetics of drugs in aquatic animals.[Method] Four formulations (NP,NB,NN and NL) of norfloxacin (NFX) in Anguilla japonica were studied by a single oral dose of 30 mg/kg NFX.The content of NFX in plasma of eels was determined by UPLC-MS and computed by non-compartment model.[Result] Pharmacokinetics parameters of NP,NB,NN and NL showed that Cmax were 1.273,1.073,0.616 and 0.094 mg/L after single drug administration 3 or 4 h;T1/2 were 15.267,24.887,33.041 and 25.891 h respectively;AUC of NP was the maximum,being 22.670 mg/(L·h).NP:NB:NN:NL=100%:99%:63%:13%.[Conclusion]Different formulation ways of drugs had greater effects on the absorption and bioavailability of medicine in aquatic animals.
