摘要

[目的]建立一种测定藜麦中总皂苷的分光光度法,研究藜麦中总皂苷的含量.[方法]采用超声波提取、香草醛-高氯酸分光光度法,以藜麦皂苷A为对照品,检测波长482 nm,样品提取采用60%乙醇溶液,料液比1:30(g:mL),提取时间30 min,提取2次,回收乙醇后用水饱和的正丁醇萃取3次.[结果]藜麦中总皂苷浓度在0.02112~0.06336 mg/mL范围内具有良好的线性关系,线性方程为A=12.169C-0.0714,相关系数0.9999.[结论]建立了超声波提取-分光光度法测定藜麦中总皂苷含量的新方法,该方法简便、准确,可用于藜麦加工工艺的评价.

Abstract

[Objective] To establish a method for determination of Quimoa saponins by spectrophotometry,and study the content of total saponins in quinoa. [Method] By ultrasonic extraction and vanillin-perchloric acid spectrophotometry,with quinoa saponins A as the reference substance, detection wavelength was 482 nm,the samples extracted with 60% ethanol solution,solid-liquid ratio was 1:30 ,extraction time was 30 minutes, twice extraction,water-saturated n-butanol extracted three times after the recovery of ethanol . [Result] The concentration of total saponins in qui-noa has a good linear relationship in 0. 02112 ~ 0. 06336 mg/mL. The linear equation was A=12. 169C-0. 0714,and the correlation coeffi-cient was 0. 9999. [Conclusion] A new method was established for determination of content of total saponins in quinoa by ultrasonic extraction and Spectrophotometry. The method is convenient and exact. It can be used for the evaluation of processing techniques of quinoa.
