摘要

[目的]对GRAS认证的枯草芽孢杆菌发酵所得N-乙酰神经氨酸进行分离纯化,获得安全性高的目的产物.[方法]通过沸水浴破壁、菌液分离、活性炭脱色、乙醇分步沉淀除杂、乙酸结晶以及重结晶等工艺对枯草芽孢杆菌基因工程菌株经培养所得的N-乙酰神经氨酸发酵液进行分离纯化.[结果]分离纯化后,N-乙酰神经氨酸纯度在96.0%以上.[结论]研究结果为N-乙酰神经氨酸应用于食品、保健品等领域提供了质量保障.

Abstract

[Objective] The aim was to separate and purify of N-acetylneuraminic acid from Bacillus subtilis which has a Generally Recognized as Safe (GRAS) status to get the safe and good production.[Method] After the fermentation of Bacillus subtilis in right medium,N-acetylneuraminic acid,was obtained by eradicating cell wall in boiling water,separating impurity,decolorization process,ethanol precipitation,acetic acid crystalli-zation,recrystallization and other steps.[Result] Through the high performance liquid chromatography (HPLC) detection,the purity of N-acetyl-neuraminic acid reached above 96.0%.[Conclusion] The results provide reference for the application of N-acetylneuraminic acid in health prod-ucts and other fields.
