椰子乙酰CoA羧化酶(ACC)基因的鉴定及表达分析

摘要
[目的]克隆椰子的乙酰CoA羧化酶(ACC)基因.[方法]通过已发表的椰子转录组注释结果,鉴定潜在的椰子ACC基因.分析ACC基因在椰子果肉和嫩叶混合样转录组中的RPKM值,同时,采用实时定量PCR技术研究5个高RPKM值的ACC基因在椰子果肉发育的不同时期的表达情况.[结果]共获得21个椰子的ACC基因,这21个ACC基因在椰子果肉和嫩叶混合样品中有较高的表达量,4个ACC基因(Unigene7806、Unigene47155、Unigene7847和CL185.Contig1)在椰子果肉发育9个月时表达量最高.[结论]该研究为解析椰子脂肪酸积累的分子机制提供了前期工作基础.
Abstract
[Objective] To clone acetyl CoA carboxylase ( ACC) genes in Cocos nucifera. [ Method] Based on the coconut transcriptome data, to identify potential coconut ACC genes. We examined the RPKM value of ACC genes in coconut leaf and fruit transcriptome. Real-time quanti-tative RT-PCR was applied to test expression level of 5 ACC genes in different developmental stages of coconut fruit, which had highest RPKM value in coconut fruit. [ Result] A total of 21 coconut ACC genes were identified. The 21 ACC genes showed high expression level in a mixture sample from coconut pulp. 4 ACC genes(Unigene7806,Unigene47155,Unigene7847 and CL185. Contig1) showed the highest expression dur-ing the development of the coconut pulp for 9 months. [ Conclusion] These study provided basic work for elucidating the molecular mechanism of fatty acid biosynthesis of coconut.
