Optimization of Sporulation Medium and Culture Conditions of the Pathogen of Cyclamen Wilt Disease
In order to obtain a high concentration spore suspension of the pathogen causing Cyclamen wilt disease, different nutrient media were preliminarily screened, and then orthogonal experiments were conducted to obtain the optimal nutrient ratio based on the initial screened nutrients. Furthermore, single factor experiments were conducted to screen the pH and culture temperature of the media. The results showed that potatoes, mung beans, and sucrose were the optimal nutritional components for the sporulation culture medium of the pathogen of Cycla-men wilt disease. The optimal ratio was mung bean 5.0%, sucrose 0.5%, and potato 20%. The optimal pH of the culture medium was 6, and the optimal culture temperature was 30 ℃. Clarifying the spore production conditions of the pathogen causing Cyclamen wilt disease lays the foundation for the research on the pathogenic mechanism, prevention and control techniques, and spore germination inhibition of Cyclamen wilt disease. At the same time, it is of great significance for guiding scientific management and comprehensive prevention and control of diseases in actual production.
