Whole Gene Sequence Analysis of a Novel Goose Astrovirus
In order to study the genetic and evolutionary characteristics of a novel goose astrovirus(N-GAstV),the full-length genes of Anhui strain AstV/AHHX were cloned in this study,and it was 7 251 nt.Sequencing analysis showed that AstV/AHHX had typical characteristics of N-GAstV.The phylogenetic tree showed that AstV/AHHX was relatively close to GDZJ37 isolates from China,G538 and G576 isolates from Shandong Province in recent years.The homology of ORF2 with GDZJ37,G538 and G576 was 100%,and the homology of ORF1a and ORF1b was more than 99.6%.Presumably they evolved from the same strain.Amino acid sequence analysis of ORF2 revealed 12 highly variable sites.They were the amino acid sites,36(Y/S/H),60(V/I),224(T/A),228(A/S/T),229(P/Q),289(T/A),376(T/A),456(D/E),540(Q/L),608(S/T),610(E/D/G),614(N/A/T).Amino acid mutations at position 224,228,229,608 and 614 may lead to changes in the confor-mation and function of viral proteins,then the pathogenic ability of the virus was altered.
