Morphological Observation and Ploidy Identification of 4 Mutant Strains of Rehmannia chingii
In the process of collecting and breeding Rehmannia for more than three years,four single plants with apparent morphological varia-tion were found,numbered as T-1,T-2,T-3 and T-4,respectively.The plant morphological indexes were measured and the results showed that the differences of morphological indexes were obvious compared with the original species.The main stem of T-1 was obviously shortened.The stigma of T-2 was exposed to the petals and the length-to-width ratio of the leaves was smaller.The stigma of T-3 was exposed to the petals and the main stem was obviously shortened.The single leaf area of T-4 was increased by nearly 3 times and the main stem was significantly short-ened.In order to clarify the chromosome ploidy of the mutant plants,the flow cytometry was used for ploidy identification.The results showed that T-1,T-2 and T-3 were identified as diploid,but T-4 was identified as tetraploid.
