固相柱净化-高效液相色谱-串联质谱法测定辣椒红色素中的苏丹红

[目的]建立一种高效液相色谱-串联质谱(HPLC-MS/MS)同时测定辣椒红色素中4 种苏丹红染料的方法.[方法]辣椒红色素样品经正己烷提取,分子印迹柱净化,高效液相色谱柱ZORABX SB-Aq C18(3.0 mm×100 mm,3.5 μm)分离,乙腈-0.1%甲酸水梯度洗脱,ESI+电喷雾扫描,多反应监测(MRM)模式下对辣椒红色素中苏丹红进行测定,外标法定量.[结果]4 种苏丹红染料在 10~200 ng/mL均呈现良好的线性关系,检出限为5 μg/kg,定量限为10 μg/kg,加标量为10、20、50 μg/kg(n=6)时,平均回收率为 71.0%~119.0%,相对标准偏差为1.53%~6.18%.[结论]该方法适用于辣椒红色素中4种苏丹红染料的定性定量分析.
Determination of Sudan Red in Capsicum Pigment by HPLC-MS/MS
[Objective]A high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)method was developed for the simultaneous determination of four Sudan red dyes in capsicum red pigment.[Method]The capsaicin pigment samples were extracted by n-hexane,purified by a special molecular imprinting column for Sudan red,separated by RP-HPLC on ZORABX SB-Aq C18(3.0 mm×100 mm,3.5 μm),water gradient elution of acetonitrile-0.1%formic acid,ESI+electrospray scanning.Sudan red in capsicum pigment was deter-mined by multiple reaction monitoring(MRM)and quantified by external standard method.[Result]The four Sudan red dyes showed a good linear relationship in the concentration range of 10.0-200 ng/mL,the detection limit was 5 μg/kg,and the quantification limit was 10 μg/kg.The average recovery was 71.0%-119.0%and the relative standard deviation was 1.53%-6.18%when the scalars were 10 μg/kg,20 μg/kg and 50 μg/kg(n=6).[Conclusion]This method is suitable for the qualitative and quantitative analysis of four Sudan red dyes in chili red pigment.
