[目的]鉴定南方红豆杉抗肿瘤内生菌HDSW,并初步研究其抗肿瘤机制.[方法]基于16S rDNA测序鉴定菌株HDSW的种属,通过MTT试验、平板细胞集落形成试验、细胞黏附试验、划痕试验和流式细胞术检测HDSW发酵液的有机溶剂提取物对胃癌细胞MGC-803的抑制活性,利用实时定量PCR方法探索其抗肿瘤的分子机制.[结果]菌株HDSW被鉴定为芽孢杆菌属(Bacillus pumilus),其发酵液的乙酸乙酯提取物作用于胃癌细胞MGC-803可有效降低其存活率,减弱其黏附能力,抑制迁移能力,诱导细胞凋亡.菌株HDSW发酵液的乙酸乙酯提取物可显著增强促细胞凋亡基因p53、Bax和Caspase 3的表达水平,抑制细胞凋亡拮抗基因Bcl-2的表达.[结论]南方红豆杉内生菌HDSW可产生具有显著抗胃癌细胞MGC-803活性的次级代谢产物,且乙酸乙酯可将其提取出来,具有潜在的开发价值.
Antitumor Activity of an Endophytic Bacteria HDSW from Taxus chinensis var.mairei Cheng et L.K.Fu
[Objective]To identify the antitumor endophytic bacterium HDSW of Taxus chinensis var.mairei and preliminarily study its antitu-mor mechanism.[Method]The strain HDSW was identified based on 16S rDNA sequencing. The inhibitory activity of the organic solvent ex-tract of HDSW fermentation broth on gastric cancer cell line MGC-803 was detected by MTT assay,plate cell colony formation assay,cell adhe-sion assay,scratch assay,and flow cytometry.The molecular mechanism was explored by real-time PCR.[Result]The strain HDSW was identi-fied as Bacillus pumilus.The ethyl acetate extract of the fermentation broth could effectively reduce the survival rate,weaken the adhesion capa-bility,inhibit migration capability,and induce cell apoptosis of the gastric cells MGC-803.The ethyl acetate extract from the fermentation broth of strain HDSW could significantly enhance the expression of pro-apoptotic genes p53,Bax and Caspase 3,and inhibit the expression of anti-ap-optotic gene Bcl-2.[Conclusion]The endophytic Bacillus HDSW from Taxus chinensis var.mairei can produce secondary metabolites with sig-nificant anti-gastric cancer cell MGC-803 activity,which can be extracted by ethyl acetate,and possess potential development value.
