摘要

为了解鼓槌石斛、细茎石斛和球花石斛种子无菌播种快繁技术,以3 种石斛种子为研究对象,探讨不同培养基、激素种类以及浓度配比对3种石斛种子无菌萌发、原球茎增殖和种苗壮苗生根的影响.结果表明:3 种石斛种子的萌发率最高均在 1/2MS基础培养基和添加10%马铃薯的1/2MS培养基上;鼓槌石斛和球花石斛原球茎增殖培养基以1/2MS+10%马铃薯+0.5 mg/L 6-BA+0.2 mg/L NAA最佳,细茎石斛原球茎增殖培养基以 1/2MS+10%马铃薯+1.0 mg/L 6-BA+0.2 mg/L NAA最佳;鼓槌石斛生根壮苗培养基以 1/2MS+0.10 mg/L IBA+10%马铃薯+2.0%蔗糖+0.6%琼脂最佳,球花石斛和细茎石斛的生根壮苗培养基以1/2MS+0.50 mg/L IBA+10%马铃薯+2.0%蔗糖+0.6%琼脂最佳.

Abstract

To understand the Dendrbium chrysotoxum,Dendrobium moniliforme and Dendrobium thyrsiflorum seed sterile plant rapid propaga-tion techniques,in this paper,three kinds of Dendrobium seeds as the research object,discusses different types of medium,hormone and con-centration ratio of 3 kinds of Dendrobium sterile seed germination,protocorm proliferation and seedling seedling rooting effect.The results showed that the highest germination rate of the 3 Dendrobium seeds was on 1/2MS base culture and 1/2MS medium with 10%potato.Dendrobi-um chrysotoxum and Dendrobium thyrsiflorum were optimized with 1/2MS+10%potato+0.50 mg/L6-BA+0.2 mg/L NAA,while Dendrobium moniliforme was optimized with 1/2MS+10%potato+1.0 mg/L 6-BA+0.2 mg/L NAA.The best rooting and seedling medium of Dendrobium chrysotoxum moniliforme was 1/2MS+0.10 mg/L IBA+10%potato+2.0%sucrose+0.6%AGAR,and the best rooting and seedling medium of Dendrobium thyrsiflorum and Dendrobium moniliforme was 1/2MS+0.50 mg/L IBA+10%potato+2.0%sucrose+0.6%AGAR.
