Bioinformatics Analysis of OLR1 Gene and Its Expression in Gonadal Axis of Guizhou Black Goat and Qianbei Ma Goat
[Objective]To study the relationship between OLR1 gene and the reproductive rate of Guizhou black goat and Qianbei Ma goat,and to provide reference for studying the molecular mechanism of OLR1 gene regulating goat follicular development.[Method]The physico-chemical properties,subcellular localization,signal peptide sites,and advanced structure of OLR1 protein were predicted by bioinformatics methods.The phylogenetic tree was constructed by homology analysis of OLR1 amino acid sequences of different species.Then RT-qPCR was used to detect the expression of OLR1 gene in the tissues of the gonadal axis of single-lamb and multi-lamb Guizhou black goats and Qianbei Ma goats.[Result]OLR1 gene encoded 275 amino acids,and the theoretical pi value was 6.37.OLR1 was an unstable hydrophilic protein without signal peptide,which was mainly composed of α-helix curl.Compared with other animals,goat OLR1 has the closest genetic distance to ruminant sheep and cattle.The results of qPCR showed that OLR1 gene was expressed in hypothalamus,oviduct,pituitary,uterus and ovary tissues of Guizhou black goat and Qianbei Ma goat.The expression in hypothalamus,uterus,ovary and pituitary of Guizhou black goat in sin-gle-lamb group was significantly higher than that in multi-lamb group(P<0.01).The expression levels in hypothalamus,pituitary and ovary of Qianbei Ma goat in single lamb group were significantly higher than those in multiple lamb group(P<0.01).[Conclusion]The expression level of OLR1 gene was negatively correlated with the reproductive traits of Guizhou black goats and Qianbei Ma goats,which could be used as an important candidate gene affecting the reproductive performance of Guizhou local goats.
