Optimization of Deep Eutectic Solvents Extraction of Total Saponins from Pfaffia glomerata by Box-Behnken Response Surface Meth-odology
[Objective]Response surface methodology was used to optimize the extraction process of total saponins from Pfaffia glomerata,providing reference for its further development and utilization.[Method]Taking amaranthaceous plant Pfaffia glomerata as the research object,total saponins were extracted using ultrasound assisted deep eutectic solvent method.Through single factor experiments,the molar ratio of urea to choline chloride,water content,liquid-solid ratio and extraction time were investigated,the total saponin yield was used as the evaluation in-dex,and the Box-Behnken response surface methodology was used to optimize the extraction process of total saponins from Pfaffia glomerata.[Result]The optimal process for ultrasound-assisted deep eutectic solvents extraction of total saponins from Pfaffia glomerata was established with the following parameters:urea to choline chloride ratio of 1∶1,water content of 38%,liquid-material ratio of 18∶1(mL∶g),and ex-traction time of 37 minutes.Under the conditions,the total saponin yield achieved was 6.26%.[Conclusion]This method has high extraction efficiency,good repeatability and simple operation,and can be used for the extraction of total saponins from Pfaffia glomerata.
