韭菜珠芽高效离体再生体系的建立

以田间发现的韭菜植株花托上部特异性芽体(称其为珠芽)为外植体,对其进行组织培养,通过对不同激素配比的研究,建立韭菜珠芽的再生体系.结果表明,使用 10%次氯酸钠溶液消毒 7 min,消毒效果最佳,无菌率可达 82.32%,MS+1.5 mg/L 6-BA+0.4 mg/L 2,4-D为最佳不定芽诱导培养基,其不定芽诱导率为78.52%,MS+2.0 mg/L 6-BA+0.5 mg/L NAA为最佳不定芽伸长培养基,不定芽伸长率可达86.63%,1/2MS+0.5 mg/L IBA为最佳生根培养基,生根频率为88.20%,其移栽成活率可达95%以上.
Establishment of an Efficient in vitro Regeneration System of Bulbel of Chinese chive
In this study,the specific bud on the upper part of the receptacle of Chinese chive plant(called bulbil)found in the field was used as the explant for tissue culture.Through the study of different hormone ratio,the regeneration system of Chinese chive bulbil was established.The results showed that using 10%sodium hypochlorite solution for 7 min had the best disinfection effect and the sterility rate was 82.32%,MS+1.5 mg/L 6-BA+0.4 mg/L 2,4-D was the best medium for adventitious bud induction,and the adventitious bud induction rate was 78.52%.MS+2.0 mg/L 6-BA+0.5 mg/L NAA was the best medium for adventitious bud elongation,and the elongation of adventitious bud reached 86.63%,and 1/2MS+0.5 mg/L IBA was the best rooting medium,the rooting frequency was 88.20%,the survival rate of transplanting was more than 95%.
