长白山山兰组织培养技术研究

[目的]研究山兰组织培养条件.[方法]以长白山山兰(Oreorchis patens)的假鳞茎为研究材料,分析消毒处理、激素组合、生根培养等对山兰假鳞茎组织培养的影响,筛选最佳消毒处理组合和假鳞茎不同阶段的最适培养基.[结果]山兰假鳞茎最适消毒处理为75%乙醇45 s+0.1%升汞表面消毒 5 min,无菌苗污染率为 10.94%,成活率为 87.62%;山兰最佳的增殖、分化和生根培养基均为 1/4 MS+1.5 mg/L ZT+0.050 mg/L NAA+20 g/L蔗糖+8g/L琼脂,pH 5.6,增殖率为78.89%,分化率为88.34%,生根率为83.85%.[结论]该研究建立了山兰的组织培养再生体系,可为长白山区其他濒危植物的保护和繁育提供技术支撑与开发基础.
Research on Tissue Culture Technology of Oreorchis patens in Changbai Mountain
[Objective]To study the tissue culture conditions of Oreorchis patens.[Method]The pseudobulbs of Oreorchis patens in Changbai Mountain were used as research materials to analyze the effects of disinfection treatment,hormone combination and rooting culture on the tissue culture of pseudobulbs of Oreorchis patens in order to screen the best disinfection treatment combination and the optimal medium for different stages of pseudobulbs.[Result]The optimal disinfection treatment of pseudobulbs was 75%alcohol for 45 s+0.1%mercuric chloride for 5 min,the contamination rate of aseptic seedlings was 10.94%,and the survival rate was 87.62%.The best medium for proliferation,differentia-tion and rooting was1/4 MS+1.5 mg/L ZT+0.050 mg/L NAA+20 g/L sucrose+8 g/L agar,pH 5.6,and the proliferation rate was 78.89%,the differentiation rate was 88.34%,and the rooting rate was 83.85%.[Conclusion]This study established the tissue culture and regen-eration system of Oreorchis patens,which can provide technical support and development basis for the protection and breeding of other endan-gered plants in Changbai Mountain area.
