为了筛选能够提高羊免疫应答的Toll样受体激动剂,设计针对羊TLR1~TLR10 基因序列的引物,采用PCR方法检测羊体内Toll样受体的表达情况.然后,将人载脂蛋白A1(ApoA1)和HMT13佐剂配伍不同的TLR1~TLR9样受体激动剂,分组免疫湖羊后采集血样,并通过琼脂扩散试验检测抗体效价.结果表明:在湖羊的血液中均可检测到TLR1~TLR10的特异性目的片段.经过3、5、7次免疫后,添加TLR3激动剂的组效价最高,说明TLR3激动剂Ploy(I∶C)免疫增强效果最佳,可作为提高羊疫苗免疫的候选激动剂.
Selecting Toll-like Receptor Agonists for the Preparation of Sheep Anti-human Apolipoprotein A1 Antibody
In order to select Toll-like agonists that can enhance the immune response in sheep,PCR method was used to design the primers for the gene sequences of TLR1-TLR10 for sheep,and detect the expression of Toll-like receptors in sheep.Subsequently,human apolipopro-tein A1(ApoA1)and HMT13 adjuvant were combined with various commercialized TLR1-TLR9 agonists.Post immunization of Hu Sheep in different groups,blood samples were collected and antibody titers were detected by using agar diffusion test.The results indicated the specific target fragments of TLR1-TLR10 were detected in the blood of Hu Sheep.After 3,5,and 7 rounds of immunization,the titer in TLR3 agonist group was the highest,which suggested that the TLR3 agonist Poly(I∶C)exhibited the best immuno-enhancing effect,so it could be used as a candidate agonist for improving the vaccine immunogenicity in sheep.
