摘要

[目的]为鉴定造成湛江某养殖场斜带石斑鱼大量死亡的病原,对皮肤滋生白色隆起疥疮的患病斜带石斑鱼进行解剖与细菌分离与纯化.[方法]通过生化反应、16S rRNA基因克隆与测序,构建系统进化树鉴定分离纯化获得的优势菌S1.采用OD600和平板活菌计数绘制V.harveyi S1生长曲线,以R2 判断其生长曲线的相关性和可信度.[结果]鉴定结果显示S1 为哈维氏弧菌,同V.harveyi hq-V149聚为一支.药敏试验结果显示,斜带石斑鱼源的哈维氏弧菌S1对诺氟沙星、头孢氨苄、左氧氟沙星、环丙沙星等12 种抗生素敏感,对青霉素G、氨曲南、链霉素、头孢唑林、头孢哌酮等12种抗生素不敏感.[结论]哈维氏弧菌S1 是造成湛江某养殖场斜带石斑鱼大量死亡的病原,其指数生长时间为5~12 h,随后进入平台期.

Abstract

[Objective]To identify the pathogen that caused deaths of Epinephelus coioides in a farm in Zhanjiang,conducted the dissection of Epinephelus coioides with white raised scabies on skin,and bacterial isolation and purification.[Method]Though biochemical reactions,16S rRNA gene cloning and sequencing,a phylogenetic tree was constructed to identify the dominant strain S1.The growth curve of V.harveyi S1 was plotted by OD600 and plate viable count.According to R2,it would determine the correlation and reliability of its growth curve.[Result]The iden-tification result showed that S1 is one of Vibrio harveyi,the same as V.harveyi hq-V149 converged into one branch.The drug sensitivity test re-sults showed that Vibrio harveyi S1 was sensitive to 12 antibiotics such as norfloxacin,cephalexin,levofloxacin and ciprofloxacin,but not to 12 antibiotics such as penicillin G,amtreonam,streptomycin,cefazolin and cefoperazone.[Conclusion]Vibrio harveyi S1 is the pathogen which caused deaths of Epinephelus coioides in a farm in Zhanjiang.The exponential growth time of S1 is 5-12 hours,and then enters the plateau peri-od.
