基于表型性状和ISSR分析的5个核桃品种亲缘关系鉴定

[目的]选择四川省栽培面积最大、产量最多的核桃品种"盐源早"(Juglans regia"Yanyuanzao"),探索"盐源早"的亲缘关系.[方法]选择"盐源早"、"三台泡核桃"(Juglans sigillata'Santai')、"漾濞泡核桃"(Juglans sigillata"Yangbi")、"温 185"(Juglans regia"Wen185")、"新新 2"(Juglans regia"Xinxin2")5个核桃品种为试验材料,测定单果重、果仁重、出仁率、坚果三径、果形指数、种壳厚以及果仁成分(粗蛋白、粗脂肪)等指标,通过方差、相关性、聚类与主成分分析等探索 5 个核桃品种的表型性状和果仁成分多样性;利用ISSR分子标记技术探究"盐源早"等5个核桃品种的遗传多样性和遗传关系.[结果]果实的表型性状与果仁成分多样性分析结果表明,5个核桃品种的单果重、果仁重、出仁率、侧径、横径、纵径、果形指数、种壳厚及果仁成分(粗脂肪、粗蛋白含量)之间差异极显著或显著(P<0.01或P<0.05).聚类分析和主成分分析结果表明,可将5个核桃品种聚为三大类群,第Ⅰ类为"温 185",第Ⅱ类为"新新 2""盐源早""漾濞泡核桃",第Ⅲ类为"三台泡核桃".利用ISSR分子标记筛选出10 条图谱条带清晰,背景明亮,重复性强,多态性较好,稳定性强的引物.通过对扩增图谱进行多态性分析结果表明,5 个核桃品种的有效等位基因数均值为 1.337 0,Nei's基因多样性指数均值为0.193 5,Shannon信息指数均值为0.285 8.聚类分析和主成分分析表明,"盐源早"和"漾濞泡核桃"聚为一类,表明亲缘关系表现较近,且相同地区的"温 185"和"新新 2"聚为一类,"三台泡核桃"聚为一类.[结论]通过对5个核桃品种的果实表型性状、果仁成分及ISSR分子标记的聚类分析得出,"盐源早"与"漾濞泡核桃"亲缘关系较近.
Identification of Genetic Relationships Among Five Walnut Varieties Based on Phenotypic Traits and ISSR Analysis
[Objective]Select the walnut variety Juglans regia'Yanyuanzao'with the largest cultivation area and the largest production in Si-chuan Province,and explore the genetic relationship of Juglans regia'Yanyuanzao'.[Method]This study selected five walnut varieties,Jug-lans regia'Yanyuanzao',Juglans sigillata'Santai',Juglans sigillata'Yangbi',Juglans regia'Wen185'and Juglans regia'Xinxin2'as re-search materials.The single fruit weight,kernel weight,kernel yield,nut diameter,fruit shape index,seed shell thickness,and kernel composition(crude protein)were measured,and the diversity of phenotypic characters and kernel component of five walnut cultivars were studied by the ANOVA,correlation,clustering and principal component.The genetic diversity of five walnut variety including'Yanyuanzao'were investigated by ISSR molecular marker technology.[Result]The single fruit weight,kernel weight,kernel yield,lateral diameter,transverse diameter,lon-gitudinal diameter,fruit shape index,seed shell thickness,and kernel composition(crude fat and crude protein content)of the five walnut va-rieties showed extremely significant or significant differences(P<0.01 or P<0.05).The results of cluster and principal component analysis showed that the five walnut varieties could be clustered into three major classes,with the class Ⅰ being'Wen185',the class Ⅱ being'Xinxin2'and'Yanyuanzao'and'Yangbi',and the class Ⅲ was'Santai'.10 primers with clear bands,bright background,high reproduc-ibility,good polymorphism and high stability were screened by using ISSR molecular markers.The results of polymorphism analysis showed that the mean number of effective alleles of the five walnut varieties was 1.337 0,the mean gene diversity index was 0.193 5,and the mean Shannon information index was 0.285 8.The clustering and principal component analysis showed that'Yanyuanzao'and'Yangbi'were clustered to-gether,indicating that the relatives of them were closer,and'Wen185'and'Xinxin2'were clustered together in the same area,and the'San-tai'was clustered into one category.[Conclusion]Cluster analysis of the fruit phenotypic traits,kernel components and ISSR molecular markers of five walnut varieties showed that,'Yanyuanzao'and'Yangbi'had a close genetic relationship.
