Correlation analysis between the expression of ADAMTS5 and CXCR4 in serum and the severity of knee osteoarthritis
Objective To investigate the correlation between the expression levels of serum depolymerin-like metalloproteinase 5(ADAMTS5)and chemokine receptor 4(CXCR4)in patients with knee osteoarthritis(KOA)and the severity of the disease.Methods A total of 136 KOA patients who were treated in the First Affiliated Hospital of Traditional Chinese Medicine of Chengdu Medical Col-lege from January 2021 to January 2023 were regarded as the study subjects,according to Kellgren Lawrence(KL)classification,KOA patients were grouped into mild group(46 cases),moderate group(68 cases)and severe group(22 cases);in addition,136 healthy peo-ple in the same period in our hospital were regarded as the healthy control group.Serum levels of ADAMTS5,CXCR4,interleukin-17(IL-17),tumor necrosis factor-α(TNF-α)and tissue inhibitor of matrix metalloproteinase 1(TIMP-1)were measured by enzyme-linked immunosorbent assay(ELISA);Pearson was applied to analyze the correlation between serum ADAMTS5 and CXCR4 levels and relat-ed indicators;the diagnostic value of serum ADAMTS5 and CXCR4 in patients with severe KOA was analyzed by the ROC curve.Re-sults Compared with the healthy control group,the levels of inflammatory factors IL-17,TNF-α and body mass index in KOA group were obviously increased,while the level of TIMP-1 was obviously decreased(P<0.05).The serum levels of ADAMTS5(3.63±1.15)g/L in the KOA group were significantly elevated compared to the healthy control group(2.23±0.59)g/L.The serum levels of CXCR4(302.48±53.46)ng/L in the KOA group were significantly elevated compared to the healthy control group(163.97±35.82)ng/L(P<0.05);the levels of serum ADAMTS5 and CXCR4 in patients with KOA showed a gradual increase with the aggravation of the severity of KOA(P<0.05);Pearson correlation analysis showed that serum ADAMTS5 and CXCR4 levels in KOA patients were positively correlated with IL-17,TNF-α and body mass index,and negatively correlated with TIMP-1(P<0.05);ROC curve results showed that the area un-der the curve(AUC)of serum ADAMTS5 and CXCR4 in the diagnosis of severe KOA patients was 0.89 and 0.89,respectively,the AUC of the combined diagnosis of the two was 0.98,which was better than their respective diagnosis alone(Z=2.25 and 3.06,P<0.05),with specificity of 96.49%and sensitivity of 95.45%.Conclusion The levels of serum ADAMTS5 and CXCR4 in KOA patients are ob-viously increased,the expression levels of both of them gradually increase with the aggravation of the severity of the patient's condition,and have important value in the diagnosis of severe KOA patients.
