The expression and significance of hsa_circ_0077223 in endometrioid endometrial carcinoma
Objective To investigate the expression level of hsa_circ_0077223 in endometrioid endometrial carcinoma(EEC)and its relationship with clinicopathological features,so as to provide theoretical basis for the study of high performance biomarkers in EEC.Methods Two EEC tissues and two normal endometrial tissues were selected for high-throughput sequencing analysis.P<0.05 and log2Fold Changes≥1.5 as the screening criteria,four circRNAs significantly up-regulated were selected for primer design to screen out hsa_circ_0077223,which was verified in 35 EEC tissues and normal endometrial tissues obtained from Weifang People's Hospital from July 2021 to July 2022.Bioinformatics analysis included GO enrichment analysis,downstream gene prediction of hsa_circ_0077223 po-tential binding site,and coding ability prediction of hsa_circ_0077223.Results The expression level of hsa_circ_0077223 in EEC was significantly higher than that in normal control group(1.646±1.102 vs.0.653±0.457),and the expression level of hsa_circ_0077223 was related to FIGO staging and the degree of tissue differentiation.The ROC curve showed that AUC was 0.85,95%CI(0.76,0.94),sensitivity 0.77,specificity 0.89,and the maximum approximation index 0.66,with statistically significant differences(P<0.001).CPAT analysis results showed that hsa_circ_0077223 had the potential to be translated into peptides,and circbank database predicted 49 miRNAs with binding sites of hsa_circ_0077223.Conclusions The expression level of hsa_circ_0077223 in normal tissues is signifi-cantly lower than that in EEC tissues and is closely related to the degree of differentiation and staging of EEC tissues.Hsa_circ_0077223 is expected to be a new molecular marker and an effective indicator for early screening of EEC.
