摘要

以茯苓菌核为原料,应用响应曲面法优化微波辅助提取茯苓多糖的工艺.在单因素实验基础上,通过Box-Behnken实验设计以及响应曲面分析法,研究料液比、pH值、微波功率、提取时间、提取次数对茯苓多糖提取率的影响.结果表明最佳提取工艺为:液料比28.3:1、pH6.8、微波功率548W、提取时间2.2min、提取次数2次.在此条件下,茯苓多糖提取率为3.04%,与预测值3.06%的相对误差小于5%.该法得到的提取工艺参数可靠,可为茯苓多糖工业化生产提供技术参考.

Abstract

Response surface method was used to optimize microwaves assisted extraction of polysaccharide from Poria sclerotium.On the basis of single factor experiment,Box-Behnken experiment design and response surface analysis were used to study the effects of solid-liquid ratio,pH value,microwave power,extraction time and extraction times on the extraction yield of Poria polysaccharide.The results showed that the optimal extraction process was as follows:liquid-solid ratio 28.3:1,pH6.8,microwave power 548W,extraction time 2.2min,extractiòn times 2 times.Under these conditions,the extraction yield was 3.04%,and the relative error between the extrac-tion yield and the predicted value was 3.06%was less than 5%.The extraction parameters obtained by this method are reliable and can provide technical reference for the industrial production of poria polysaccharide.
