Study on the Culture of F1 Protocorms and Rhizomes of Cymbidium and Doumbalium
The primary protocorm and rhizome of Cymbidium hybridium'Dafeng'and C.serratum var.Goeringii'Taijishengmei'F1 were used as experimental materials.The method of adding different plant hormones in 1/2MS medium was adopted.The effects of different plant hormones on the proliferation and differentiation of F1 protocorms and rhizomes of Cymbidium hybridium'Dafeng'and C.serratum var.Goeringii'Taijishengmei'were studied,in order to provide reference for efficient breeding of Cymbidium.The results showed that 1/2MS+banana 80.0 g·L-1+carbon powder 0.5 g·L-1as the basic medium,TDZ 1.0 mg·L-1 and NAA 0.2 mg·L-1 were the most beneficial to rhizomes proliferation,and the proliferation rate reached 343.33%.The addition of 6-BA 2.0 mg·L-1 and NAA 0.3 mg·L-1 in basic medium was beneficial to the differentiation of rhizomes,and the differentiation rate reached 83.33%.The optimum formula for the growth of protocorm was as follows,basic medium plus TDZ 1.0 mg·L-1 and NAA 0.2 mg·L-1,the growth rate reached 160.00%.The optimum formula for protocorm differentiation was as follows,6-BA 2.0 mg·L-1 and NAA 0.4 mg·L-1 were added in basic medium,and the differentiation rate was 76.00%.According to the anatomical observation,both protocorm and rhizomes had opaque vascular bundle centers,and the proportion of opaque vascular bundle centers in rhizome was larger than that in protocorm.In addition,the proliferation and bud differentiation rates of F1 generation rhizomes were higher than those of the original bulbs.Both rhizomes and protocorms have the characteristics of roots in their internal structures,which are composed of epidermis,cortex and vascular tissue from inside to outside.
