Community Composition and Diversity Analysis of Endophytes in Hippophae rhamnoides Seeds From Different Locations
In order to provide reference for the subsequent study of the biological activity of endophytes and explore the endophytic functional microbiota in Hippophae rhamnoides seeds,six samples were collected from Xinjiang,Inner Mongolia,Shanxi,Shaanxi and Gansu,the structure and diversity of endophytic bacteria and fungi were studied by high-throughput sequencing and bioinformatics analysis methods based on 16S rRNA and ITS2 sequence.The results showed that the composition of endophytes in H.rhamnoides seeds was relatively simple.The endophytic bacteria were mainly Proteobacteria,Firmicutes,Actinobacteria;the endophytic fungi were Ascomycete,Mortierellomycota and Basidiomycota.Alpha diversity analysis showed that the Shannon and Simpson indices of endophytic bacteria in Zhidan,Shaanxi and Altay,Xinjiang samples were significantly different.Meanwhile,the Shannon and Chaol indices of endophytic fungi were significantly different between seed samples from Lyuliang,Shanxi and Zhidan,Shaanxi.Non-metric multidimensional scaling analysis showed certain similarity on the composition of endophytes in different seed samples.Kyoto Encyclopedia of Genes and Genomes analysis generated six primary metabolic systems and 26 secondary metabolic pathways in the endophytic bacteria.Accordingly,the diversity and community composition of endophytes in H.rhamnoides seeds from different regions were different,which may be closely related to their growing environments.
