寒地特色薄皮甜瓜转录组测序分析

以13份薄皮甜瓜为试材,采用转录组测序方法,研究了白粉病抗性相关基因的表达情况,以期获得薄皮甜瓜白粉病抗性相关候选基因.结果表明:上调表达谱中,有21个基因只出现在抗性较好的品种中;下调表达谱中,有42个基因只出现在低抗品种中.GO分析显示,基因产物的活动主要涉及碳-碳裂解酶活性、ADP焦磷酸水解酶活性、肌氨酸氧化酶活性和β-麦芽糖4-α-葡萄糖转移酶活性等.利用KEGG Pathway分析了解基因的生物学功能发现,上调和下调基因多集中于植物昼夜节律调节、鞘糖脂生物合成通路硫胺素代谢和细胞的包吞作用等.结合前期GWAS的分析结果,选定MELO3C033293.2和ME-LO3C006706.2等作为寒地薄皮甜瓜白粉病抗性相关候选基因,进行下一步定量表达分析.
Transcriptome Sequencing Analysis of Oriental Melon in Cold Region
The expression of powdery mildew resistance-related genes in 13 oriental melon (Cucumis melo L.)cultivars was studied by transcriptome sequencing,in order to obtain candidate genes for powdery mildew resistance.The results showed that there were 21 genes in up-regulated expression profile only in resistant cultivars and 42 genes in down-regulated expression profile only in low-resistant cultivars.The GO analysis showed that the activities of the gene products were mainly related to the activities of carbon-carbon lyase,ADP pyrohydrolase,sarcosine oxidase and β-maltose 4-α-glucosyltransferase.By analyzing the biological functions of genes in KEGG Pathway,it was found that the up-regulated and down-regulated genes were mainly involved in plant circadian regulation,thiamine metabolism in glycosphingolipid biosynthesis pathway and endocytosis of cells.Based on the results of GWAS analysis,MELO3C033293.2 and MELO3C006706.2 were selected as candidate genes related to powdery mildew resistance of oriental melon in cold region,proceed with the next step of quantitative expression analysis.
