Transcriptome Sequencing Analysis of Oriental Melon in Cold Region
The expression of powdery mildew resistance-related genes in 13 oriental melon (Cucumis melo L.)cultivars was studied by transcriptome sequencing,in order to obtain candidate genes for powdery mildew resistance.The results showed that there were 21 genes in up-regulated expression profile only in resistant cultivars and 42 genes in down-regulated expression profile only in low-resistant cultivars.The GO analysis showed that the activities of the gene products were mainly related to the activities of carbon-carbon lyase,ADP pyrohydrolase,sarcosine oxidase and β-maltose 4-α-glucosyltransferase.By analyzing the biological functions of genes in KEGG Pathway,it was found that the up-regulated and down-regulated genes were mainly involved in plant circadian regulation,thiamine metabolism in glycosphingolipid biosynthesis pathway and endocytosis of cells.Based on the results of GWAS analysis,MELO3C033293.2 and MELO3C006706.2 were selected as candidate genes related to powdery mildew resistance of oriental melon in cold region,proceed with the next step of quantitative expression analysis.
