Investigation and Pathogen Identification of Watermelon Virus Disease in Typical Hilly Area of South Taihang
The field occurrence and symptoms of watermelon virus disease were used as test materials and were investigated in the open field and small arch shed planted in the typical hilly area of South Taihang,and the pathogen of watermelon virus disease was studied by small RNA sequencing and RT-PCR,in order to provide reference for the development of watermelon virus disease prevention and control strategy in this area.The results showed that the watermelon virus disease in the field generally started from early to middle of May,and the symptoms were mainly mosaic and wrinkling.A total of 7 viruses were detected in 14 disease samples using 16 virus primers.They were watermelon mosaic virus (WMV ),zucchini yellow mosaic virus (ZYMV ),papaya malformed mosaic virus (PLDMV ),melon aphid yeltization virus (MABYV ),watermelon ruffle associated virus 2 (WCLaV2),watermelon crypto-virus (CiLCV)and melon mixed virus 1 (CMAV1 ).The detection rate of WMV,CiLCV and CMAV1 virus was 100%,and the detection rate of MABYV virus was the lowest (28.6%).In the field,watermelon virus disease occurred in complex infection,and the disease samples were generally infected with 4-7 different kinds of viruses.Further consistency comparison and analysis of the full-length genomic nucleotide sequence of WMV showed that WMV isolates from this region had high genetic diversity,indicating that the virus may have multiple initial infection sources in this region.
