观赏用微型兰花组织培养最佳配方的优化

以大花蕙兰'大凤'×豆瓣兰'太极圣梅'(Cymbidium hybridium'Dafeng'×C.serratum var.goeringii'Taijishengmei')F1代试管苗为试材,采用在培养基中添加不同的有机物质,筛选最适生长的培养基;并在此基础上,在培养基中添加不同色素,研究其对植株生长的影响及观赏效果,以期为兰花微型培养的观赏应用研究提供参考依据.结果表明:在1/2 MS+IBA 0.5 mg·L-1+6-BA 2 mg·L-1+碳粉0.5 g·L-1的培养基中,添加75 mL·L-1椰汁有利于兰花试管苗叶长、叶宽、株高以及鲜质量的增加,对植株生长效果最好;在1/2 MS+IBA 0.5 mg·L-1+6-BA 2 mg·L-1+碳粉0.5 g·L-1+椰汁75 mL·L-1的培养基中添加不同浓度的食用色素红曲红、叶绿素铜钠盐、栀子蓝,经121℃高温灭菌后培养基的颜色均会发生变化.叶绿素铜钠盐浓度为533.3 mg·L-1的彩色培养基植株生长良好,培养基整体观赏效果最佳.
Optimization of the Best Formula for Tissue Culture of Ornamental Miniature Orchid
The F1 test-tube seedlings of Cymbidium hybridium 'Dafeng'×C.serratum var.goeringii'Taijishengmei'were used as the test materials.Different organic substances were added to the medium to screen the optimal growth medium.On this basis,different pigments were added to the medium to study their effects on plant growth and ornamental effects,in order to provide reference for the study of ornamental application of orchid microculture.The results showed that in the medium of 1/2MS+IBA 0.5 mg·L-1+6-BA 2 mg·L-1+carbon powder 0.5 g·L-1,added 75 mL·L-1 coconut juice was benefited to the increased of leaf length,leaf width,plant height and fresh weight of orchid plantlets in vitro,and had the best effect on plant growth.In 1/2MS+IBA 0.5 mg·L-1+6-BA 2 mg·L-1+carbon powder 0.5 g·L-1+coconut juice 75 mL·L-1 medium added different concentrations of edible pigment monascus red,sodium copper chlorophyllin,gardenia blue,after 121 ℃ high temperature sterilization medium color was changed.The plants grew well in the color medium with the concentration of sodium copper chlorophyllin of 533.3 mg·L-1,and the overall ornamental effect of the medium was the best.
