Establishment and Optimization of Tissue Culture System of Four Hosta plantagina Varieties
Taking the young shoot explants of four Hosta plantagina varieties as the test materials,the effects of different sterilisation times,phytohormone ratios and concentrations on the induction,proliferation and rooting of adventitious shoots,as well as the effects of different substrate formulations on seedling transplantation were investigated,in order to provide reference for enriching the resources of Hosta plantagina in the Northeast China,and for enriching the market demand as well as the factory production of Hosta plantagina.The results showed that the optimal sterilization method for explants was 75%alcohol 30 seconds+0.1%mercuric chloride solution for 8 minutes.The optimal formulations of clumping bud induction medium were H.'Indigo Leaf'MS+6-BA 3.5 mg·L-1+NAA 0.1 mg·L-1,H.'Golden Tiara'MS+6-BA 2.5 mg·L-1+NAA 0.2 mg·L-1,H.'Flos Lotus'and H.'Blue Cadet'MS+6-BA 2.5 mg·L-1+NAA 0.1 mg·L-1;H.'Flos Lotus'and H.'Blue Cadet'showed high value-added of 4.06 and 6.71 times,respectively,under the induction of MS+6-BA 3.0 mg·L-1+NAA 0.1 mg·L-1,'Golden Tiara'showed 4.23 times under the induction of MS+6-BA 4.0 mg·L-1+NAA 0.1 mg·L-1.H.'Indigo Leaf'could proliferate 4.25-fold under the induction of MS+6-BA 2.0 mg·L-1+NAA 0.2 mg·L-1,and the optimal rooting medium was H.'Indigo Leaf'1/2MS+IBA 0.5 mg·L-1,H.'Golden Tiara'and H.'Blue Cadet'1/2MS+NAA 0.05 mg·L-1,and H.'Flos Lotus',1/2MS+NAA 0.1 mg·L-1.In the seedling refining test,the most suitable substrate for transplanting H.'Flos Lotus'and H.'Blue Cadet'tissue culture seedlings was the substrate of upper sand and lower soil(thickness 1∶1),and the survival rate was 100%.The H.'Indigo Leaf'and H.'Golden Tiara'were transplanted with sand∶soil(1∶1)mixed substrate for transplanting.
