Effect of GA3 and 6-BA on Breaking Dormancy and Promoting Germination of Rose Seeds
The seeds of low-maintenance rose variety'Glory'after soaking with different concentrations of GA3 and 6-BA were used as experimental materials.The contents of endogenous hormones GA,ABA,IAA and CTK in rose seeds treated with 0,50,100,150 mg·L-1 and 200 mg·L-1GA3 and 6-BA and the relative expression levels of hormone-related key genes were determined by enzyme-linked immunosorbent assay(ELISA)after 90 days of low temperature stratification(refrigerator 4 ℃).The effects of different concentrations of GA3 and 6-BA on breaking dormancy and promoting germination of'Glory'seeds were studied,in order to provide reference for screening the optimal hormone concentration to break the dormancy of rose seeds.The results showed that there were significant differences in germination potential and germination rate of rose seeds treated with different concentrations of GA3 and 6-BA,and the germination rates of seeds soaked with 150 mg·L-1GA3 and 50 mg·L-1 6-BA were the highest(55.56%and 46.67%).Compared with the control,the content of ABA in rose seeds after GA3 and 6-BA treatment was decreased,while the contents of GA,IAA and CTK were significantly increased.The ratio of GA+IAA+CTK/ABA in different concentrations of GA3 and 6-BA treatment was significantly higher than that in the control,and the ratio was the highest under 150 mg·L-1GA3 and 100 mg·L-1 6-BA treatment,indicated that the ratio of GA+IAA+CTK/ABA could be increased by adjusting GA3 or 6-BA soaking.The optimal concentration was 150 mg·L-1 GA3 or 50-100 mg·L-1 6-BA.The key genes of hormone synthesis in rose seeds showed different changes under hormone treatment,which may achieve the effect of antagonizing ABA and promoting seed germination by down-regulating the transcription of GA2ox and ABI5 and up-regulating the transcription of ABI4 and WRKY13.
