Isolation and Identification of Mycoparasites of Chinese Rose Powdery Mildew
Taking the leaves of Chinese rose powdery mildew as the test materials,the mycoparasite were isolated by the tissue separation method,and their parasitic ability was determined according to the reconnection test.Combined with morphological characteristics and rDNA-ITS sequence determination,the species of mycoparasite were identified,in order to provide a reference for disease prevention and control.The results showed that 13 strains of mycoparasite were isolated from the diseased leaves with typical symptoms.After the representative strains KMR3 and KMR12 with strong parasitic ability parasitised the capsule shell,the cell wall of powdery mildew spores was broken and the contents were released.The colony of strain KMR3 was white at the beginning,and then gradually developed into brown colony.The conidia were brown,oval,long rod,oval or inverted pear-shaped.ITS sequence was 99%similar to Alternaria alternata(MT482506.1)and Alternaria arborescens(PP 097783.1).In the phylogenetic tree,the sequence of strain KMR3 was the closest to Alternaria alternata(MT482506.1)and Alternaria arborescens(PP 097783.1).The colony of strain KMR12 was white at the initial stage,and gradually developed into brown or light yellow colony at the later stage.The conidia were brown or dark brown,oval,rod-shaped or inverted pear-shaped,and ITS sequence was 100%similar to those of Alternaria angustifoidea(MK 910070.1)and Alternaria longipes(MK675101.1).In the phylogenetic tree,the sequence of strain KMR12 was the closest to that of Alternaria angustifoidea(MK 910070.1)and Alternaria longipes(MK675101.1).Using morphological characteristics and ITS sequence analysis,strains KMR3 and KMR12 were identified as Alternaria.
