Taking Taxus chinensis as the experiment material,based on the full-length transcriptome data of T.chinensis,with the application of bioinformatics methods,TcDof family genes were screened and identified,the physical and chemical properties,conservative domain structure,phylogenetic tree,and the expression profiles of the TcDof genes in tissue regeneration after bark girdling were analyzed,in order to provide reference for further analysis of the biological function of TcDofs genes in T.chinensis.The results showed that the total of 39 Dofs contained open reading frame(ORF).The coding region of TcDofs were 534-1 599 bp in length,their numbers of amino acid were between 177 aa and 532 aa,their isoelectric point were between 5.45 and 9.81,all TcDofs location were in the cell nucleus.The TcDofs had a typical C2-C2 single zinc finger structure.Phylogenetic tree analysis found that Taxus chinensis could be divided into A,C,D three subfamily containing 8 conserved motifs,and all TcDofs proteins had the Motif 1 conserved motif.The expression levels of TcDof1,TcDof38,and TcDof26 genes were significantly upregulated during the bark regeneration process of Taxus chinensis,suggesting that they regulate the bark tissue regeneration of Taxus chinensis.
