Schisandrin B Improves Blood Lipid Levels in Hyperlipidemic Mice by Modulating Gut Microbiota
Objective Preliminary exploration of the effect of Schisandrin B on lipid metabolism in hyperlipi-demic mice from the perspective of gut microbiota.Methods Select 90 male C57BL/6 mice and randomly divide them into blank control group,model group,Schisandra total lignan group(50 mg/kg),Schisandra B low-dose group(2 mg/kg),Schisandrin B medium-dose group(4 mg/kg),and Schisandrin B high-dose group(8 mg/kg).Except for control group,other groups were fed with high-fat diet(HFD)for 12 weeks.At week 8,each group was orally administered treatments.The control group and model group were given the same volume of solvent for 4 consecutive weeks.Two mouse feces samples were collected in a sterile environment one day before the end of administration.Fasting for 12 hours after administration,blood and liver samples were taken,and the levels of TG,TC,HDL-C,and LDL-C in serum were detected using reagent kit;HE staining method was used to observe the pathological morphological changes in liver;and 16S rRNA sequencing technology was used to study the regulatory effects of gut microbiota structure and function in each groups of mice.Results Compared with control group,the mice in model group showed a significant increase in body weight,and serum levels of TG,TC,and LDL-C were significantly increased,while HDL-C levels were significantly reduced(all P<0.01).The liver showed mild hepatic steatoid lesions.Compared with model group,the mice in drug administration group showed a significant decrease in body weight,a significant decrease in serum levels of TG,TC,and LDL-C,and a significant increase in HDL-C levels(all P<0.05,P<0.01).The 16S rRNA sequencing results showed that,compared with control group,the composition of gut microbiota in model group mice exhibited significant differences in multiple phyla.Compared with model group,the intervention of Schisandrin total lignan group and Schisandrin B improved the similarity of OTU(operational taxonomic units)composition in the gut microbiota of hyperlipidemic mice,reduced the ratio of Firmicutes/Bacteroidetes(F/B)in the gut microbiota of hyperlipidemic mice,promoted the abundance of beneficial bacteria such as Candidatus_Saccharimonas,Muribaculaceae,Clostridia_UCG-014,Lach-nospiraceae_NK4A136_group,also inhibited the growth of the harmful bacteria Escherichia shigella.Conclusion Schisandrin B can significantly regulate serum TC,TG,HDL-C,and LDL-C levels in hyperlipidemic mice,improve liver steatosis,and its mechanism of regulating blood lipids may be achieved by regulating the gut microbiota of hyperlipidemic mice.
