The Development of Reference Intervals of Serum Alkaline Phosphatase for Healthy Children Aged 0 to 12 Years in Nanning,Guangxi Based on a Laboratory Information System
Objective To investigate the distribution of the dataset of serum alkaline phosphatase (ALP)from Laboratory Information System (LIS)and to establish the reference intervals of serum ALP for healthy children aged 0-12 years in Nanning,Guangxi,so as to provide a reliable reference basis for clinical diagnosis and treatment.Methods A total of 2314 healthy children aged 0 to 12 years in Nanning area who visited the International Zhuang Medicine Hospital Affiliated to Guangxi University of Traditional Chinese Medicine for physical examination were selected as the reference population,while another 240 cases were collected for the reference interval validation group.All study subjects were divided into 6 age groups:<28d, 28 d to <1 year,1 year to <3 years,3 years to <6 years,6 years to <9 years,and 9 years to <12 years. Kolmogorov-Smirnov Z test was used to analyze the distribution of the dataset after removing outliers using the interquartile range method.The Mann-Whitney U test was used to analyze the differences in ALP levels between sex and adjacent age groups,an Spearman's rank correlation was used to investigate the correlation between ALP levels and age groups.The non-parametric method (P2.5 to P97.5 )was used to establish serum ALP reference intervals.Results After excluding outliers and duplicates,a total of 2256 cases were included in the study cohort,and the dataset showed a skewed distribution.There was no statistically significant difference in ALP levels between sexes (P>0.05),while a positive correlation between ALP levels and age (r=0.526,P<0.05)was observed.Age groups with no statistically significant differences were combined,and the reference intervals of serum ALP which had been verified were 105.98-288.03 U/L (<28 d),136.60-350.40 U/L (28 d to <3 years),and 153.00-354.10 U/L (3 years to <12 years), respectively.There were certain differences compared with the reference intervals provided in the instructions and industry standards.Conclusion The current study establishs reference intervals for serum ALP in healthy children aged 0 to 12 years,which is important for improving the application value of serum ALP testing in children in clinical laboratories in this area.
